Specimen Requirements

Container/tube:  
1.    Collect on culture transport swab containing Amies or Stuart’s media.   Eswabs are accepted.  Order a fungal culture for sources other than vaginal or throat.  
2.    Label with two patient identifiers (full name, DOB), date and time of collection and specimen source.

Collection Instructions:
1.    Throat:  
 Obtain by rubbing whitish patches of exudate if present.
2.    Vaginal:  Before collecting form mucosal membrane of vagina, wipe away excessive amounts of secretion and discharge.  If a bacterial culture and Gram stain are also requested, collect a second swab to maximize test sensitivity.

Specimen Transport:  Maintain at ambient temperature and transport to laboratory within 24 hours.
 

Specimen Rejection:   Swabs received in viral transport media.

Methodology

Specimens are plated to Sabouraud dextrose agar and to BHI blood agar which contains chloramphenicol and gentamicin to inhibit bacterial contamination.  Cultures are incubated at 35C for 3 days.  Identification of yeast to genus and species level is performed upon request.
