Specimen Requirements

Specimen must arrive within 30 minutes of collection. If
this is not possible, the specimen may be refrigerated for up to 2
hours, however, the specimen must be received in the laboratory
within 2 hours of collection or the results may be adversely
affected due to deterioration of the specimen.

 

Container/Tube: VACUTAINER® plastic,
screw-top urine cup (kit supplied)

Specimen Volume: 10 mL from a clean-catch,
midstream or catheterized urine collection,

Collection Instructions: Note type of
collection.

1. The first-morning void is preferable.

2. Clean-catch urines for culture should be collected with the
VACUTAINER® urine collection kits supplied.

3. Unscrew the cap of the urine cup. Place the cap on the
counter, top down. Do not contaminate interior of cap or cup.

4. Cleanse the patient yourself or instruct the patient to
collect a clean-catch, midstream specimen as follows:

 

Males

A. Wipe head of penis in a single motion with the first
Towelette®.

B. Repeat with the second Towelette®.

C. If not circumcised, hold the foreskin back before
cleansing.

D. Urinate a small amount into the toilet or bedpan and proceed
to step #5.

 

Females

A. Separate the labia.

B. Wipe the inner folds front to back in a single motion with
the first Towelette®.

C. Wipe down through center of labial folds with the second
Towelette®.

D. Keep the labia separated, urinate a small amount into the
toilet or bedpan, and proceed to step #5.

 

5. Place the cup under the stream and continue to urinate into
the cup and collect specimen.

6. Finish voiding into toilet or bedpan.

7. Place the cap on the cup and tighten
securely
. Give specimen to nurse. Label container with
patient’s name (first and last), date and time of collection,
initials of person collecting the specimen, and type of specimen.
Submit this specimen for urinalysis testing.

8. If a #UC “Culture, Urine” or a
#CIND “Culture, Urine, if Indicated” has been
ordered in addition, collect the grey-top (lyophilized urine
maintenance formula) VACUTAINER® tube from kit as
follows:

A. Do not handle the tube by the cap.

B. Remove the paper cover from the filled cup to expose the well
containing the needle and push the grey-top (lyophilized urine
maintenance formula) covered tube, cap downward, onto the
needle.

C. After the tube has filled, remove it from the needle, shake
the tube well.

D. If a small amount of urine is collected (<10 mL), remove
the lid to the specimen cup. Remove the grey stopper from the
culture tube, and pour urine into the tube cup to at least the
minimum fill line (5.0 mL) and mix well.

E. Label tube with patient’s name (first and last), date
and actual time of collection, patient number, and initial’s
of person collecting the specimen.

F. Submit the grey-top (lyophilized urine maintenance formula)
VACUTAINER® tube along with the urine cup.

Note:  The grey-top (lyophilized urine
maintenance formula) VACUTAINER® tube cannot be used for
routine urinalysis testing.

9. Upon delivery of specimen to Boulder Community Hospital
Laboratory, specimen must be signed in. STAT specimens must be
signed in and handed to a technologist.

10. Please submit routine specimens from 6 a.m. to 8:30 p.m.
unless the patient is scheduled for surgery the next day or has
trouble voiding.

Color: straw to dark yellow

Appearance: clear to cloudy

Specific gravity: 1.002-1.030

pH: 4.6-8.0

Urobilinogen: 0.2-1.0 EU

Protein: negative

Glucose: negative

Ketones: negative

Bilirubin: negative

Blood: negative

Reducing substances

Negative

Critical Value: Positive ketones, glucose or reducing substances
if <1 year of age

Nitrite: negative

Leukocyte esterase: negative

Microscopic: a small amount of WBCs, RBCs, epithelial
cells, hyaline casts, and mucus

Note: 1. When the results of the urine
macroscopic is negative, a microscopic is not indicated and will
not be performed. A microscopic will be performed if the protein,
blood; glucose, blood; leukocyte esterase; or nitrite is
positive.

2. If the physician has specifically ordered a microscopic,
order #UMIC “Urine Microscopic Only, Urine.”
